EXTRACTION OF AMYLASE USING DIFFERENT PLANT LEAVES AS SUBSTRATE

Abstract

Enzymes are biological molecules that speeds up the reaction in a significant way. Fermentation plays an important role in the production of all kind of enzyme. Submerged fermentation holds a vital for the production of enzyme amylase using Aspergillus niger. In this work, different solid substrates such as Prosopis juliflora, Calotropis gigantea, Lucas aspera leaves were used as substrate for the production of amylase as they are very cheap and also easily available raw materials. Aspergillus niger was identified to be the best producer of amylase and they are also available naturally. Amylase that was produced using all the three substrates was having good activity but the maximum enzyme activity was found in Prosopis juliflora  (0.95U and specific activity of 19U/mg of protein).
